Assessment of the association between SMAD1 and HHIP gene variation and non-syndromic cleft-lip palate in Chilean case-parent trios Genet. Mol. Biol.
Suazo,José; Santos,José Luis; Jara,Lilian; Blanco,Rafael.
Nonsyndromic cleft lip/palate (NSCLP) is a congenital malformation with features of a complex genetic trait. Several studies have reported positive association and linkage between NSCLP and microsatellite markers in the 4q28-4q33 region particularly with the D4S192 (4q31) marker. We hypothesized that the candidate genes SMAD1 and HHIP (4q31) could be involved in the etiology of NSCLP based on previous positive linkage results and their important role in maxillofacial development. We evaluated the possible association between microsatellite markers located at less than 1 cM from these genes and NSCLP using a sample of 58 Chilean case-parent trios. Microsatellite markers were analyzed using the polymerase chain reaction (PCR) with fluorescent labeled...

Association between G316A growth hormone polymorphism and economic traits in pigs Genet. Mol. Biol.
Faria,Danielle Assis de; Guimarães,Simone Eliza Facioni; Lopes,Paulo Sávio; Pires,Aldrin Vieira; Paiva,Samuel Rezende; Sollero,Bruna Pena; Wenceslau,Amauri Arias.
The association between G316A growth hormone polymorphism and quantitative traits was investigated in an F2 population of pigs. Association analyses were performed using a statistical model that included genotype, sex, batch and sex by genotype interaction as fixed effects and sire as random effect. The polymorphism was associated with the number of right teats (p = 0.03), heart weight (p = 0.04), lung weight (p = 0.05), carcass length determined by the Brazilian carcass classification method (p = 0.04), picnic shoulder weight (p = 0.07), jowl weight (p = 0.01), pH 24 h after slaughtering (p = 0.03) and drip loss (p = 0.01). Interaction between genotype and sex was observed for six performance traits. The additive effect was significant (p < 0.10) for...

Can survival of European flat oysters following experimental infection with Bonamia ostreae be predicted using QTLs? ArchiMer
Harrang, Estelle; Heurtebise, Serge; Faury, Nicole; Robert, Maeva; Arzul, Isabelle; Lapegue, Sylvie.
The present study identifies quantitative trait loci (QTLs) in response to an experimental infection with the parasite responsible for bonamiosis, Bonamia ostreae, in two segregating families of the European flat oyster, Ostrea edulis. We first constructed a genetic-linkage map for each studied family and improved the existing genetic-linkage map for the European flat oyster with a set of SNP markers. This latter map now combines the best accuracy and the best estimate of the genome coverage available for an oyster species. Secondly, by comparing the QTLs detected in this study with those previously published for O. edulis in similar experimental conditions, we identified several potential QTLs that were identical between the different families, and also...

Candidate mutations used to aid the prediction of genetic merit for female reproductive traits in tropical beef cattle R. Bras. Zootec.
Fortes,Marina Rufino Salinas; Enculescu,Charmaine; Porto Neto,Laercio R.; Lehnert,Sigrid A.; McCulloch,Russell; Hayes,Ben.
ABSTRACT In this study, we aimed to provide a wet laboratory validation for a set of single nucleotide polymorphisms (SNP), which had been identified as candidate functional variants in silico. Genotyping for candidate SNP was performed in Brahman and Tropical Composite cattle. After quality control, 29 SNP were first investigated individually for their association with female reproductive traits and then used as a panel for genomic predictions. The reproductive traits studied were age at first corpus luteum (AGECL; days), post-partum anoestrus interval (PPAI; days), and a binary trait that described if the cow had ovulated before weaning the first calf or not (PW, 0-1). Single nucleotide polymorphisms in six genes (FOXA2, TRAF4, IRF2, IRF1, BPTF, and...

Combining six genome scan methods to detect candidate genes to salinity in the Mediterranean striped red mullet (Mullus surmuletus) ArchiMer
Dalongeville, Alicia; Benestan, Laura; Mouillot, David; Lobreaux, Stephane; Manel, Stephanie.
Background: Adaptive genomics may help predicting how a species will respond to future environmental changes. Genomic signatures of local adaptation in marine organisms are often driven by environmental selective agents impacting the physiology of organisms. With one of the highest salinity level, the Mediterranean Sea provides an excellent model to investigate adaptive genomic divergence underlying salinity adaptation. In the present study, we combined six genome scan methods to detect potential genomic signal of selection in the striped red mullet (Mullus surmuletus) populations distributed across a wide salinity gradient. We then blasted these outlier sequences on published fish genomic resources in order to identify relevant potential candidate genes...

Expression of VINV1, SCB1 and SCS1 genes in the seed coats of contrasting soybean seeds1 Rev. Ciênc. Agron.
Bahry,Carlos André; Zimmer,Paulo Dejalma.
The seed coat is vital for the proper development of soybean seeds and also, after their physiological maturity, for providing protection against various biotic and abiotic factors. The aim of this work was to evaluate the relative expression of three candidate genes, VINV1, SCB1 and SCS1, which are possibly involved in seed quality, in the contrasting seed coats of four soybean genotypes. Two yellow seed coat genotypes, BMX Potência RR and CD 202, and two black seed coat genotypes, TP and IAC, were used. Relative gene expression was evaluated by the qPCR technique for seven phases of seed coat development in the four genotypes, at 25, 30, 35, 40, 45, 50 and 55 days after anthesis. The design was completely randomised, with three replications. Data were...

Abstract Drought limits wheat production in the Brazilian Cerrado biome. In order to search for candidate genes associated to the response to water deficit, we analyzed the gene expression profiles, under severe drought stress, in roots and leaves of the cultivar MGS1 Aliança, a well-adapted cultivar to the Cerrado. A set of 4,422 candidate genes was found in roots and leaves. The number of down-regulated transcripts in roots was higher than the up-regulated transcripts, while the opposite occurred in leaves. The number of common transcripts between the two tissues was 1,249, while 2,124 were specific to roots and 1,049 specific to leaves. Quantitative RT-PCR analysis revealed a 0.78 correlation with the expression data. The candidate genes were...

Genetics of coffee quality Braz. J. Plant Physiol.
Leroy,Thierry; Ribeyre,Fabienne; Bertrand,Benóit; Charmetant,Pierre; Dufour,Magali; Montagnon,Christophe; Marraccini,Pierre; Pot,David.
Coffee quality, in the present context of overproduction worldwide, has to be considered as a main selection criterion for coffee improvement. After a definition of quality, and an overview of the non genetic factors affecting its variation, this review focuses on the genetic factors involved in the control of coffee quality variation. Regarding the complexity of this trait, the different types of quality are first presented. Then, the great variation within and between coffee species is underlined, mainly for biochemical compounds related to quality (caffeine, sugars, chlorogenic acids, lipids). The ways for breeding quality traits for cultivated species, Coffea arabica and Coffea canephora are discussed, with specific challenges for each species. For C....

Influence of temperament-related genes on live weight traits of Charolais cows R. Bras. Zootec.
Garza-Brenner,Estela; Sifuentes-Rincón,Ana María; Rodríguez-Almeida,Felipe Alonso; Randel,Ronald D.; Parra-Bracamonte,Gaspar Manuel; Arellano-Vera,Williams.
Abstract We investigated whether a panel of molecular markers previously associated with temperament has an effect on live growth traits. Phenotypic data from 412 Charolais cows categorized according to their age in adult and young cows were used to determine Pearson's correlations between birth (BW), weaning (WW), and yearling live (YW) weight and temperament traits (measured as exit velocity [EV] and temperament score [TS]). For association analysis, selective genotyping of a group of 80 cows identified as the most docile and temperamental were genotyped with a 151-SNP (single-nucleotide polymorphisms) panel of molecular markers previously associated with temperament. Significant Pearson's correlations between birth weight and weaning weight and the two...

LINKAGE MAPPING OF CANDIDATE GENES FOR INDUCE RESISTANCE AND GROWTH PROMOTION BY Trichoderma koningiopsis (Th003) IN TOMATO Solanum lycopersicum Acta biol.Colomb.
SIMBAQUEBA,JAIME; COTES,ALBA MARINA; BARRERO,LUZ STELLA.
Induced systemic resistance (ISR) is a mechanism by which plants enhance defenses against any stress condition. ISR and growth promotion are enhanced when tomato (Solanum lycopersicum) is inoculated with several strains of Trichoderma ssp. This study aims to genetically map tomato candidate genes involved in ISR and growth promotion induced by the Colombian native isolate Trichoderma koningiopsis Th003. Forty-nine candidate genes previously identified on tomato plants treated with Th003 and T. hamatum T382 strains were evaluated for polymorphisms and 16 of them were integrated on the highly saturated genetic linkage map named “TOMATO EXPEN 2000”. The location of six unigenes was similar to the location of resistance gene analogs (RGAs), defense related...

Polymorphism in Litopenaeus vannamei genes and cross-species amplification in other shrimp species PAB
Santos,Camilla Alves; Farias,Flavio; Teixeira,Ana Karina; Rocha,João Luís; Guerrelhas,Ana Carolina; Freitas,Patrícia Domingues de.
Abstract: The objective of this work was to assess polymorphisms within Litopenaeus vannamei candidate genes related to growth performance, and a possible association between weight genotypes and phenotypes in two shrimp closed lines. Three EST-SSR (serine-arginine, 60S ribosomal, and troponin) and one EST-SNP (crustacyanin) were selected and evaluated in L. vannamei and seven other marine shrimp species, besides two freshwater species. Genotypes had no effect on total body weight. Although no association was observed in the sampled population, these markers showed amplification for most of the evaluated species and, therefore, may be useful in further genetic studies on shrimp species.

The Brazilian contribution to Attention-Deficit/Hyperactivity Disorder molecular genetics in children and adolescents Genet. Mol. Biol.
Genro,Júlia Pasqualini; Roman,Tatiana; Rohde,Luis Augusto; Hutz,Mara Helena.
Attention-Deficit/Hyperactivity Disorder (ADHD) is a common psychiatric condition of children worldwide. This disorder is defined by a combination of symptoms of inattention and hyperactivity/impulsivity. Diagnosis is based on a sufficient number of symptoms causing impairment in these two domains determining several problems in personal and academic life. Although genetic and environmental factors are important in ADHD etiology, how these factors influence the brain and consequently behavior is still under debate. It seems to be consensus that a fronto-subcortical dysfunction is responsible, at least in part, for the ADHD phenotype spectrum. The main results from association and pharmacogenetic studies performed in Brazil are discussed. The investigations...
